【預期用途】

表皮細胞生長因子受體（EGFR）是人表皮生長因子受體（HER）家族成員，屬于受體酪氨酸激酶（TKI）家族，為原癌基因c-erbB1表達產物。EGFR主要位于細胞膜，被EGF等配體激活后發生二聚化并引發胞內段產生酪氨酸激酶活性，進一步激活下游信號轉導，細胞生長、增殖等多種生理過程。

本試劑盒用于惡性腫瘤（肺癌(ai)、結直腸癌(ai)、胰腺癌(ai)、鼻咽癌(ai)等(deng)實體(ti)(ti)瘤）患(huan)者組織樣(yang)本中的Kras基因12和13密碼子突變的定性檢測，為(wei)個(ge)體化用藥治療提供依據。Kras基因突變與惡性腫瘤患者EGFR信號傳導通路的靶向藥物原發性耐藥有關(guan)。Kras基因野生型的患者能從愛必妥 (Erbitux，又稱西妥昔單抗/Cetuximab)或維克替比(Vectibix，又稱帕尼單抗/Panitumumab)、易(yi)瑞沙或特羅(luo)凱的治療中獲益，而KRAS基因突變的惡性腫瘤(liu)患者無明顯受(shou)益于(yu)此類靶向藥物(wu)治(zhi)療(liao)。

本試劑盒可檢測出如下KRAS基因7種(zhong)突變：

12密碼子的GGT>AGT、GGT>CGT、GGT>TGT、GGT>GAT、GGT>GCT、GGT>GTT  6種突變和13密碼子的GGC>GAC。
